Gel Casting Instructions

Ensure that the gel solution is fully prepared and that the glass plates are clean and dry.

1. Assembly
- Place the gel casting clamp on a flat, stable surface.
- Fully open the pressure plate on the clamp (Pressure Plate – Position 1).
- Insert the flat glass plate and the concave (notched) glass plate as shown in the reference figure.
- Make sure the bottoms of both glass plates are properly aligned.
- Rotate the pressure plate to secure and tighten the glass plates (Pressure Plate – Position 2).

a. Pouring the Resolving Gel (Lower Gel)

After pouring the resolving gel, liquid sealing, surface leveling, and waiting for solidification are not required.
You may directly pour the stacking gel (upper gel) solution and allow the gel to polymerize.

The gel casting stand may be used individually or combined into a larger unit as needed, enabling efficient use of laboratory bench space.
Before beginning gel casting, confirm the following:
- The flat and concave glass plates are vertically and horizontally aligned.
- The bottom edges of the glass plates are correctly positioned.
- The pressure plates at both ends are firmly tightened.
2. Gel Casting
a. Pouring the Resolving Gel (Lower Gel)
- Press the lower edge of the assembled glass plates onto the silicone base.
- Secure the upper edge of the concave glass plate with a clip to hold the plates firmly in place.

Slowly pour the prepared resolving gel solution into the chamber between the two glass plates until it reaches the scale line.
- Servicebio glass plates include scale markings for accurate pouring.
- For glass plates without scale lines, pour the solution to approximately 1.5–2.0 cm below the upper edge of the flat glass plate.
b. Using the Servicebio One-Step Gel Preparation Kit (G2177)
After pouring the resolving gel, liquid sealing, surface leveling, and waiting for solidification are not required.
You may directly pour the stacking gel (upper gel) solution and allow the gel to polymerize.
c. Optional: Liquid Sealing Step
- Carefully add distilled water or absolute ethanol along the sides of the gel chamber to seal and level the resolving gel surface.
- Allow the gel to stand for approximately 20 minutes until solidified.
- Remove the sealing liquid and absorb any residual moisture with absorbent paper.
- Slowly pour the prepared stacking gel solution until the chamber is full.
- Immediately insert the electrophoresis comb.
- Allow 15–20 minutes for complete polymerization.
