Operation Steps

Before starting, ensure that the electrophoresis tank, gel tray, sample comb, gel maker, and all related components are clean and dry.

1. Preparation
Weigh an appropriate amount of agarose into an Erlenmeyer flask and add the required volume of buffer. Heat the mixture in a water bath, on a magnetic heating stirrer, or in a microwave until the agarose is completely melted. Gently swirl to ensure a uniform agarose solution.

2. Gel Casting
Place the gel maker horizontally on the laboratory bench.
Select a suitable gel tray based on experimental needs (available tray sizes: 120 × 120 mm, 120 × 60 mm, 60 × 120 mm, and 60 × 60 mm).
Position the gel tray in the gel maker—two gels can be prepared simultaneously using 60 × 60 mm trays; other sizes allow for one gel at a time.
Insert the sample comb securely into the designated slot.
Add 5 μL of SerRed (or another nucleic acid dye) to the agarose solution cooled to approximately 55°C. Mix gently and pour the solution into the gel tray, spreading it evenly across the surface.
Allow the gel to solidify completely at room temperature, then carefully remove the sample comb vertically to complete gel preparation.

3. Sample Loading
Transfer the gel (with tray) to the electrophoresis tank, positioning the sample wells near the negative electrode (black).
Add running buffer until the gel is fully submerged.
Using a 10 μL micropipette, load samples into the wells, changing the pipette tip after each sample to prevent cross-contamination.
Avoid damaging the gel surface around the wells and follow the correct sample loading order.

4. Electrophoresis
Attach the lid, ensuring the correct polarity (red = positive, black = negative).
Connect the power leads to the electrophoresis power supply and set an appropriate voltage based on experimental requirements.
During electrophoresis, samples migrate from the negative (black) to the positive (red) electrode.
Stop electrophoresis when the bromophenol blue dye front is approximately 1 cm from the lower edge of the gel.

5. Observation
When paired with Servicebio’s blue light module and compatible power supplies (e.g., BE-SPW-DES Dual Module Programmable Power Supply or BE-SPW-6S Power Supply), electrophoresis progress can be monitored in real time, allowing for optimized separation and improved experimental outcomes.

Safety and Maintenance Precautions

1. Electrical Safety
Do not operate the unit while it is connected to power without the top cover securely in place.
Always ensure the lid of the horizontal electrophoresis apparatus is closed before switching on the power supply.
Do not touch the apparatus during electrophoresis operation to prevent the risk of electric shock.

2. Cleaning and Maintenance
All components of this product can be cleaned with water. After washing, allow parts to air dry naturally or wipe dry with absorbent paper.
Do not dry at high temperatures or expose the components to direct heat sources, as this may cause deformation or damage to the product.

Operation Steps

Before beginning, ensure that the electrophoresis tank, gel tray, sample comb, gel maker, and all related components are clean and dry.

1. Preparation
Weigh an appropriate amount of agarose into an Erlenmeyer flask and add the required volume of buffer.
Heat the mixture in a water bath, on a magnetic stirrer with heating, or in a microwave oven until the agarose is fully melted.

Gently swirl to ensure uniform mixing and obtain a clear agarose gel solution.

2. Gel Casting
Place the gel maker horizontally on the lab bench.

Select a suitable gel tray according to experimental needs (available tray sizes: 120×120 mm, 120×60 mm, 60×120 mm, and 60×60 mm).
Insert the gel tray into the gel maker — two 60×60 mm gels can be cast simultaneously; other sizes accommodate one gel at a time.
Position the sample comb securely in place.

Add 5 μL of SerRed (or another nucleic acid dye) to the agarose solution cooled to approximately 55°C. Mix gently and pour evenly into the gel tray.

Allow the gel to solidify completely at room temperature, then carefully remove the sample comb vertically to form sample wells.

3. Sample Loading
Transfer the gel (with tray) into the electrophoresis tank, positioning the sample wells close to the negative electrode (black).

Add the running buffer until the gel is fully submerged.

Using a 10 μL micropipette, load samples into each well. Replace the pipette tip after each sample to prevent cross-contamination.

Avoid damaging the gel surface and maintain a consistent loading order.

4. Electrophoresis
Secure the top cover, ensuring proper alignment of the positive (red) and negative (black) electrodes.

Connect the leads to the electrophoresis power supply and set the desired voltage according to experimental parameters.

During electrophoresis, the samples migrate from the negative (black) to the positive (red) electrode.

Stop the run when the bromophenol blue dye front is approximately 1 cm from the lower edge of the gel.

5. Observation
The BE-SVL-2 Integrated Horizontal Electrophoresis System can be equipped with a blue light transilluminator, allowing real-time observation of electrophoresis progress when used with the SPW-6S electrophoresis power supply.
